Born in Odessa and educated in St. Petersburg, Milstein formed a duo with Vladimir Horowitz in 1923. After touring the newly-founded Soviet Union for two years, they went on a tour abroad and decided not to return. Later they formed a trio with Grigory Piatigorsky. Like the other two, Milstein eventually settled in New York and became a U.S. citizen.
Pludermacher, a graduate from the Paris Conservatory, was Milstein's regular accompanist at the time. He also made himself a name as a performer of contemporary music.
